Optical method for studying DNA-protein interactions at the single-molecule level

Abstract

We describe a single-molecule method for studying DNA based on tethered particle motion, a gold nano-bead, total internal reflection and adequate biophysical analysis. Using the system we measured the DNA three-dimensional distribution and dynamics.
